The DNA damage response - from cell biology to human disease

نویسندگان

چکیده

Maintenance of DNA integrity is crucial for faithful transmission the genetic code from generation to generation. Our constantly under attack both endogenous and exogenous sources damage. To ensure genome stability, cells have developed elegant damage repair mechanisms. Defects in been linked several human diseases including promoting oncogenesis, heritable neurodegenerative neuromuscular caused by unstable repeats, neuropathies myopathies mutations rearrangements mitochondrial DNA, neuropsychiatric disorders, premature aging syndromes. This review will discuss our current understanding how these underlying errors contribute clinical outcomes patients who present with diseases.

Bacterial Genotoxins: Merging the DNA Damage Response into Infection Biology

Bacterial genotoxins are unique among bacterial toxins as their molecular target is DNA. The consequence of intoxication or infection is induction of DNA breaks that, if not properly repaired, results in irreversible cell cycle arrest (senescence) or death of the target cells. At present, only three bacterial genotoxins have been identified. Two are protein toxins: the cytolethal distending tox...

The Role of chk2 in Response to DNA Damage in Cancer Cells

Accumulation of gene changes and chromosomal instability in response to cellular DNA damage lead to cancer. DNA damage induces cell cycle checkpoints pathways. Checkpoints regulate DNA replication and cell cycle progression, chromatin restructuring, and apoptosis. Checkpoint kinase 2 (chk2) is activated in response to DNA lesions. ATM phosphorylate chk2. The activated Chk2 kinase can phosphoryl...

Quantitation of genome damage and transcriptional profile of DNA damage response genes in human peripheral blood mononuclear cells exposed in vitro to low doses of neutron radiation

Background: Humans are exposed to ionizing radiation from different sources that include natural, occupational, medical, accidental exposures. Evaluation of the effect of low level of neutron exposure to human cells in vitro has important implications to human health. Attempts were made to measure genome damage, transcriptional profile of DNA damage response and repair genes in peripheral blood...

Cell-autonomous circadian DNA damage response

3718 Cell Cycle Volume 11 Issue 20 Of the hallmarks of cancer,1 the process of invasion and metastases is arguably the least well-understood and therefore least drugable. Invasion through the extracellular matrix is arguably the critical distinction between carcinoma in situ and carcinoma with metastatic potential. Efforts to understand targetable mechanisms of cancer invasiveness are sorely ne...
